Separation of hydrophilic basic analytes and analytes that are labile at low pH require the use of high pH eluents.
Silica based phases that are generally used in reversed phase LC are slowly dissolved at pH above 7, with an increasing dissolution rate with increasing pH. Three beta-blockers were separated using Nanosep Reversed Phase Nanoparticles at pH 10 (image).
With such a high pH, severe degradation of many reversed phase HPLC columns can be expected. The polymer nanoparticles are however stable also at extreme pH.
